Abstract:Objective To detect the variation of blood nitric oxide (NO), creatine kinase (CK), superoxide dismutase (SOD) and malonyldialdeyde (MDA) and the apoptosis of myocardial tissues in type 2 diabetic rats, and to explore the protective effects of folic acid and vitamin B12 on myocardial tissues in rats. Methods Sixty SD rats aged 6 weeks were used to establish rat models of diabetes mellitus successfully, and then randomly divided into the control group ( group A), the model group (group B) and folic acid and vitamin B12 intervention group (group C). After 12 weeks, all the rats were sacrificed for assaying the levels of NO, CK, SOD and MDA in the blood drawn from hearts. Tunel method was used to observe myocardial cell apoptosis. Immunohistochemical method was employed to determine the expression of caspase-3 and caspase-8, and the average gray density values of caspase-3 and caspase-8 were measured by using computer image analysis system. Results After 12 weeks, the levels of blood NO and SOD were significantly lower in group B than in group A (P<0.01), but significantly higher in group C than in group B (P<0.01). The levels of CK and MDA were significantly higher in group B than in group A (P<0.01), but significantly lower in group C than in group B (P<0.01). The apoptotic indexes of myocardial cells in cardiac muscle tissues of the 3 groups were (2.6357±0.5214),(16.2859±0.6983) and (4.0410±0.3121) respectively. The expression of Caspase-3 and Caspase-8 in the 3 groups was (79.2471±2.3547) and (70.2596±3.2118), (97.1530±4.9522) and (98.9322±5.0583), (84.1549±3.0198) and (78.3299±3.5680) respectively. The apoptotic indexes of myocardial cells and the expression of caspase-3 and caspase-8 were significantly higher in groups B and C than in group A (P<0.01), but significantly lower in group C than in group B (P<0.01). Conclusions The changes of blood NO, CK, SOD and MDA in myocardial tissues of type 2 diabetic rats may participate in the process of cardiomyocyte apoptosis. Intervention by folic acid combined with vitamin B12 can effectively alleviate the myocardial injury.
