Regulating role of hypoxia-inducible factor 1α on apoptosis of human sperm

Abstract:Objective To explore the role and mechanism of hypoxia-inducible factor 1α (HIF-1α) in regulating apoptosis of human sperm, and to provide a novel theoretical basis for diagnosis and treatment of diseases such as oligozoospermia. Methods Adult semina were divided into the normal sperm vitality group and the oligozoospermia group. Sperms in the two groups were incubated with HIF-1α inhibitor cobalt chloride or lificiguat (YC-1) respectively, and control groups without drug treatment were set up respectively. The computer-assisted semen analysis system was used to detect the vitality (expressed as progressive motility rate) of sperms in each group. Enzyme-linked immunosorbent assay was applied to measuring the sperm apoptosis level and acrosin activity. Western blotting was performed to detect the levels of HIF-1α, B-cell lymphoma-2 (Bcl-2) and Bcl2-associated X protein (Bax). Results Compared with the normal control group, the apoptosis rate and expression levels of HIF-1α and Bax in the cobalt chloride treatment group increased, but the sperm vitality, acrosin activity and expression level of Bcl-2 decreased. Compared with the oligozoospermia control group, the apoptosis level of sperms and expression level of Bax in the YC-1 treatment group decreased, but the sperm vitality, acrosin activity and expression levels of HIF-1α and Bcl-2 increased. Conclusion The level of HIF-1α can affect the sperm apoptosis and vitality.
