Abstract:Objective To investigate the changes of percentage of peripheral blood CD26+CD4+T cell and the serum level of soluble CD26(sCD26) in patients with pulmonary tuberculosis. Methods Thirty healthy donors (HD), 30 patients with susceptible primary tuberculosis (S-TB) and 30 patients with multidrug-resistant tuberculosis (MDR-TB) were enrolled in this study. 2 ml peripheral venous blood with heparin anticoagulation was stained with multicolor labeled monoclonal antibody of CD3, CD4 and CD26. Beckman flow cytometry (FCM) was used to count the percentage of CD26+CD4+T cell. ELISA was employed to determine peripheral serum level of sCD26. Results The percentage of CD26+CD4+ T cell in all CD4+ T cells was significantly lower in the MDR-TB group ((13.63±3.85)%) than in the S-TB group ((20.17±5.94)%) and HD group ((26.55±5.77)%) (F=45.037, P=0.000). The serum level of sCD26 was significantly higher in the MDR-TB group ((617.99±55.57) ug/ml) than in the HD group ((410.23±36.21) ug/ml) and S-TB group ((435.22±38.34)ug/ml) (F=197.287, P=0.000). Spearman correlation analysis showed that there was a negative correlation between the percentage of CD26+CD4+ T cell and the serum level of sCD26 in healthy donors, patients with susceptible primary tuberculosis and patients with multidrug-resistant tuberculosis (r=-0.391, P=0.002). Conclusions With the development of pulmonary tuberculosis from susceptible primary tuberculosis to multidrug-resistant tuberculosis, the percentage of peripheral blood CD26+CD4+T cell decreases gradually, while the serum level of sCD26 increases significantly, and there is a negative correlation between them. Determination of the percentage of peripheral blood CD26+CD4+ T cell and serum level of sCD26 in patients with pulmonary tuberculosis is conducive to knowing the progression of tuberculosis disease.
