Application of detecting tumor markers in colorectal tissue to the early diagnosis of colorectal cancer

Abstract:Objective To explore the differences between the concentration of tumor markers in tissue extract and serum concentration of tumor markers in colorectal cancer, and their relationships with early diagnosis and treatment of colorectal cancer. Methods We selected 40 hospitalized patients with colorectal cancer in the First Hospital of Changsha City. The patients' tumor tissues and adjacent peritumoral tissues were taken as the tumor tissue group and the adjacent peritumoral tissue group; meanwhile, benign colorectal tissues from 50 patients without colorectal cancer were selected as the control group. Cell disruption method was used to extract supernatant liquid of tissue cells, and Roche electrochemical luminescence method to determine the concentration of carcinoembryonic antigen (CEA), carbohydrate antigen 199 (CA199) and carbohydrate antigen 125 (CA125) in the supernatant. Differences in the tissue and serum concentration of CEA, CA199 and CA125 were compared, and the relationships of the above-mentioned tumor markers in colorectal cancer tissue with clinical pathological characteristics were analyzed. Results The concentration of CEA, CA199 and CA125 was significantly higher in the tumor tissue group than in the adjacent peritumoral tissue group and the control group as well as higher in the tumor tissue group than in the serum group, showing statistically significant differences (P<0.05). The ROC curve areas of CEA and CA199 in the tumor tissue group were 0.928 and 0.863, respectively, which were significantly higher than those in the serum group. The concentration of CEA and CA199 in the tumor tissue group was positively correlated with the invasion depth of colorectal cancer and lymph node metastasis, and the concentration of CEA, CA199 and CA125 in the tumor tissue group was positively correlated with tumor node metastasis (TNM) staging of colorectal cancer, displaying statistically significant differences (all P<0.05). Conclusion Detecting tumor markers CEA, CA199 and CA125 in extract of colorectal cancer tissue can be used for early auxiliary diagnosis of colorectal cancer, and it has a certain application value.
