Abstract:Objective To explore the role of berberine in regulating adiponectin and its influence on blood lipid indexes and atheromatous plaque formation in rats with hyperlipidemia(HLP). Methods Hyperlipidemia rat models were induced by feeding male Wistar rats with high-cholesterol diet for 30 days, and another 10 rats treated with regular food and water were selected as the control group. After 10 weeks, the 30 hyperlipidemia rats were randomly assigned into 3 groups, HLP group (n=10), HLP+LBB group (n=10) and HLP+HBB group (n=10). The latter two groups were respectively given low-dosage (150 mg/(kg·d)) and high-dosage (300 mg/(kg·d)) berberine for 10 weeks, while the control group and the HLP group were given normal saline. After 10 weeks, the serum levels of triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), serum total adiponetin and high molecular weight (HMW) adiponetin were tested and compared between the four groups; meanwhile, the infarction areas of the rat hearts were evaluated and compared. Results Compared with the control group, the serum levels of TG, TC and LDL-C in the HLP group were significantly increased, but the serum level of HDL-C was significantly decreased, suggesting that the HLP rat models were successfully established. Compared with the HLP group, the serum levels of TG, TC and LDL-C in the HLP+LBB group and the HLP+HBB group were significantly decreased (P<0.001), but the serum level of HDL-C in the HLP+LBB group was significantly increased (P<0.05). No statistically significant difference was found in the serum total adiponectin level among the 4 groups (P>0.05). The ratio of HMW adiponetin to total adiponetin was significantly lower in the HLP group than in the control group,but it was significantly higher in the HLP+HBB group than in the HLP group (P<0.05). Compared with the control group, the expression of AdipoR1 protein in the HLP group was down-regulated, but it was significantly higher in the HLP+HBB group than in the HLP group, with a statistically significant difference (P<0.05). The expression of AdipoR2 protein in the HLP group showed no apparent variation, but it was significantly higher in the HLP+HBB group than in the control group(P<0.05). The infarction area in the HLP+HBB group was significantly less than those in the HLP group and the HLP+LBB group ((11.4±1.0)% vs.(52.2±0.9)%, (11.4±1.0)% vs. (33.1±1.1)%, both P<0.05). Conclusions Berberine can confront the deregulation of adiponectin expression in hyperlipidemia rats, and it may be a potential therapeutic strategy for blood lipid and atheromatous plaque formation.
