Abstract:objective To investigate the effect of curcumin on the angiotensin receptor type-1 in rat heart with heart failure. Methods Models of chronic heart failure (CHF) established by left anterior descending coronary artery ligature. Thirty-two Wistar rats were randomly divided into four groups: sham operation group, sham operation with curcumin (2.0 mmol/L in drinking water) treatment group, CHF model group, CHF model with curcumin (2.0 mmol/L in drinking water) treatment group. Cardiac function was examined by echocardiography after 4 weeks therapy. PRA and AngⅡ levels were tested by radioimmunity. The mRNA levels of AT1 receptor were measured by qRT-PCR. In addition, the level of cardiac AT1 receptor expression was detected by Westernblotting. Results Compared with sham operation group, left ventricular ejection fraction (LVEF) was significantly decreased. The plasma levels of PRA and AngⅡ were markedly increased. Moreover, the AT1 receptor mRNA and protein expressions were also higher in rat heart with heart failure. All of the above-mentioned abnormalities were reversed to some degree by curcumin treatment in rats with heart failure, but not sham operation group rats. Conclusions curcumin reduces expression of AT1 receptor in heart and improves heart function in rats with heart failure.
