Abstract:ObjectiveTo preliminary investigate the 30-day toxicity of Momordica charantia extract(MCE)in BALB/c mice. MethodA total of 80 female mice weighing 18-22 g were randomly divided into two clusters based on body weight, and each weight-based cluster included fiour groups (10 mice per group) : negative control ,low, intermediate, and high dose MCE groups, Then mice were administered intragastrically with MCE at dosage of 0, 1.11, 3.33 and 10.00g/kg BW respectively for 30 days,and body weights were measured once every week to adjust the gavage volume. By the end of study, blood samples were collected for blood routine test and biochemical indexes, organ weighting and the spleen cell counting, histopathologic examinations were performed on all animals euthanized for necropsy. ResultsThe weight gain of mice in all treated groups was similar to that in non-treated group. liver/body, kideny/body, spleen/body and thymus/body coefficients of mice in all treated groups were similar to those in non-treated group(P>0.05). Hematology results showed that the level of hemoglobin in middle dosage group of MCE were significantly lower than those in the negative control group(P<0.05), the percentage of lymphocyte in high dosage group of MCE were significantly lower than those in the negative control group(P<0.05), and the percentage of monocyte in high dosage group of MCE were significantly higher than those in the negative control group(P<0.05). White blood cell count and differential count, red blood cell count, hemoglobin, hematocrit, mean corpuscular volumem and mean corpuscular hemoglobin concentration of mice in all treated groups were normal(P>0.05); Clinical biochemical parameters showed the level of glucose in the high dosage group of MCE was significantly higher than the negative control group (P<0.05), the level of creatinine in the high dosage group of MCE was significantly lower than the negative control group(P<0.05). however, all the changes in detected index of blood routine and blood biochemical were within the scope of this historical reference in our lab, so there were no biological significance. Aspartate transaminase, alanine aminotransferase, blood urea nitrogen, creatinine, cholesterol, triglyceride, glucose, albumin and serum total protein of mice in all treated groups were normal(P>0.05). No pathological changes in liver, Kidney, spleen, thymus, lymph node, and bone marrow of mice in all treated groups were observed under microscope. Conclusion No obvious toxicity effects was observed in mice fed with MCE for 30 days.
