Abstract:Objective To study the effect of resveratrol (Res) on the levels of homocysteine (Hcy) and indices of oxidative stress in aging mice induced by D-galactose(D-gal). Methods Forty-eight mice were divided into 4 groups: control group, aging model group, Res low dose (22.5mg/kg)group and Res high dose(45mg/kg) group. The mice in the control group were given daily subcutaneous injection with saline, and those in the other three groups were given daily subcutaneous injection with D-gal. Moreover, the mice in the control group and aging model group were given sodium carboxymethyl cellulose solution, and the mice in the Res low dose group and Res high dose group were given Res dissolved in sodium carboxymethyl cellulose solution. After 16 weeks, homocysteine (Hcy) level in serum, superoxide dismutase(SOD), glutathione peroxidase(GSH-Px), total antioxidant capacity(T-AOC) and malondialdehyde(MDA) levels in serum and liver of mice were measured. Results Compared with the mice in the control group, the levels of Hcy in serum and MDA in serum and liver of the mice in the aging model group were significantly increased (P<0.05), while the levels of SOD, GSH-Px and T-AOC in serum and liver were significantly decreased (P<0.05), suggesting the aging model was successful. Compared with the mice in the aging model group, administration of Res at low and high doses significantly lowered the levels of Hcy in serum and MDA in serum and liver, and raised the levels of SOD, GSH-Px and T-AOC in serum and liver. Conclusions Different doses of Res can prevent the increase of oxidative stress and Hcy induced by D-gal in mice. Res has anti-aging effect.
