摘要 目的检测自身免疫性慢性荨麻疹(AIU)患者外周血单个核细胞(PBMCs)中Toll样受体7 (TLR7) mRNA与蛋白的表达,探讨AIU的发病机制。方法采集AIU患者30例与正常对照组30例外周血,淋巴细胞分离液分离PBMCs,提取RNA,采用实时荧光定量逆转录PCR检测 TLR7 mRNA表达;采用免疫荧光染色、流式细胞仪检测PBMCs中TLR7蛋白的表达。结果与正常对照组相比,AIU患者PBMCs中TLR7 mRNA的表达无差异(p>0.05);蛋白表达增强,差异有统计学意义(p<0.05)。结论AIU患者PBMCs中TLR7蛋白表达增强,TLR7可能在AIU的发病中起作用。

Abstract :Objective To investigate the expression of TLR7 mRNA and protein in peripheral blood mononuclear cells (PBMCs) of patients with autoimmune chronic urticaria (AIU) and to explore the immunology mechanism of AIU. Methods PBMCs from 30 patients with AIU and 30 normal controls were separated by Ficoll-Hypaque Solution. The expression of TLR7 mRNA and protein were detected respectively by real-time quantitative reverse transcription polymerase chain reaction and by flow-cytometric analysis. Results The results showed that, as compared to normal controls, the expression of TLR7 protein in PBMCs of patients with AIU significantly increased (p <0.05), but there was no difference (P >0.05) in the expression of TLR7 mRNA. Conclusions The expression of TLR7 increases in patients with AIU, and the findings suggest that TLR7 may play roles in the pathogenesis of AIU.
