Abstract:Objective The aim is to explore the effects of Perfluorooctane sulphonate, (PFOS) on zebrafish embryo development of and ontents of SOD, GSH and MDA in zebrafish. Methods The health male fishes and female fishes were selected to mating and spawning. Postfertilized four hours embryos were exposed to either DMSO or various concentration of PFOS (0.1, 0.5, 1.0, 2.0, 5.0mg/L). Embryonic hatching rate, abnormality rate, heart rate and malformation were counted. Contents of SOD, GSH and MDA in 120hpf zebrafish tissue were analyzed by kits respectively. Results With the increment of PFOS concentration, the hatching rate of embryo and the heart rate of embryo were reduced, the malformation rate of embryo and the lethality of larval increased. Contents of SOD and GSH decreased, and content of MDA increased in 120hpf zebrafish. When the PFOS concentration was 5.0mg/L, the heart rate of embryo, the hatching rate of embryo, the malformation rate of embryo, and the lethality of larval was 109.3± 5.2 per minute, 21.53%, 59.17% and 60.8% respectively. PFOS exposure significantly decreased the GSH content, depressed the SOD activity, and increased level of MDA in 120hpf zebrafish tissue. Conclusion Our results indicated that oxidative stress would be involved in developmental toxicity of zebrafish.
