Effects of mitomycin C on life span, fertility and motor ability of Drosophila melanogaster

Abstract:Objective To explore the effects of different concentration of mitomycin C (MMC) on the life span, fertility and motor ability of Drosophila melanogaster. Methods Wild-type Drosophila melanogastermales and females werecultured in the media containing different concentration of MMC (0 μg/ml, 40 μg/ml, 80 μg/ml and 120 μg/ml).The median lethal time, the average life span, the highest life expectancy, numbers of offspring, eclosion rate and climbing abilityof drosophilias in each group were determined.Statistical analysis was performed and the t-test was used for comparison among groups. Results The median lethal time, the average life span, the highest life expectancy, numbers of offspring, eclosion rate and climbing index ofdrosophiliasin the 80 μg/ml and 120 μg/ml MMC groups were significantly reduced as compared with the control group (all P<0.01), and with the increase of MMC concentration, each index gradually decreased as compared with the control group. In contrast to the control group, the eclosion rate and numbers of the F3 generation drosophilias in the 40 μg/ml MMC group was significantly reduced, showing statistically significant differences (P<0.05, P<0.01). The numbers of drosophilas, life span and motor ability had no obvious differences between the F1 and F2 generations. The median lethal time of female drosophilas and the median lethal time, the average highest life expectancy and the average life span of male drosophilas were significantly shortened, and the differences were statistically significant (P<0.05). Conclusion MMC with the concentration of more than 80 μg/ml had adverse effects on the fertility, lifespan and motor ability of drosophilas.
