Abstract:Objective To establish a method for determination of phosphatidylcholine in health food by high performance liquid chromatography with evaporative light scattering detector (HPLC-ELSD). Methods Using n-hexane-isopropanol aqueous solution (0.1% triethylamine and pH 3.5 adjusted by trifluoroacetic acid) as mobile phase, phosphatidylcholine in health food samples was determined by HPLC-ELSD equipped with a silica gel column. Results In the range of 0.05 mg/ml-2.0 mg/ml, the logarithm of the response value of ELSD was linearly related to the logarithm of the concentration of phosphatidylcholine, and the detection limits were 0.02 mg/L. The recovery rates were 93.7%-102.3%, and the precision (n=7) was less than 1.3%. Conclusions The developed HPLC-ELSD method can rapidly determine the content of phosphatidylcholine in health food with the advantages of simple operation, high accuracy and good reproducibility, and it is suitable for determination ofphosphatidylcholine content in health food.
