Abstract:Objective To establish an efficient, simple and rapid method for the detection of florfenicol and florfenicol amine residues in animal-derived food by ultra high performance liquid chromatography-tandem mass spectrometry. Methods The optimization of sample pretreatment procedures as follows: the food samples were extracted by ammoniation acetonitrile and purified by C18 solid phase extraction packing. After that, the extraction was determined by ultra high performance liquid chromatography-tandem mass spectrometry and quantified by internal standard method. Results In this method, the limits of detection of florfenicol and florfenicol amine were both 0.5 μg/kg, and the limits of quantitation were both 1.5 μg/kg. A good linear relationship was obtained with the concentration ranged from 1.0 μg/L to 40.0 μg/L, and the recoveries of three different spiked concentrations were all above 75%, with the relative deviation being less than 10%. Conclusion Ammoniation acetonitrile was selected to extract the food samples in this method, which increased the sample pretreatment recoveries. This method is high-efficient, simple, accurate and reliable, which can be used for detecting florfenicol and florfenicol amine residues in animal-derived food.
