Abstract:Objective To study the optimal combination of extraction method of total flavonoids content in puerariae soft capsule. Methods The content of total flavonoids determined by ultraviolet spectrophotometry was used for evaluating the experimental effects. Polyamide powder particle size, ethanol concentration, ultrasonic power and ultrasonic temperature were employed as the experimental factors. L9(34) orthogonal experiment design was selected to screen the optimal combination. Results The optimal combination was 70% ethanol concentration, 200w power, 60℃ ultrasound and 60-80 mesh powder polyamide column chromatography. Validation test showed that the total flavonoids content was 3.49 g/100 g. Conclusions The optimal combination of extraction method of total flavonoids content in puerariae soft capsule is verified through the experiment, and it avoids the low content of total flavonoids caused by improper extraction method. The results of validation test are satisfactory.
