Effects of ginsenoside Rb1 on the expression of nerve growth factor and its receptortyrosine kinase A in rat dorsal root ganglion after peripheral nerve injury

Abstract:Objective To observe the influence of ginsenoside Rb1 on nerve growth factor (NGF) and its receptor tyrosine kinase A (TrkA) expression in dorsal root ganglion in rats with peripheral nerve injury. Methods SD rats were randomly divided into the sham operation group, the model group, the methylcobalamin group (10.8 mg/kg) and the low-dose (25 mg/kg) and high-dose (50 mg/kg) ginsenoside Rb1 groups. Nerve clamping injury method was used to establish the peripheral nerve injury models, and then the model rats were administered intragastrically for 14 successive days. After the last dose, the sensory recovery of the rats was assessed using the photothermal pain threshold method. The expression of NGF and its receptor TrkA in rat dorsal root ganglion was detected by Western-blotting, and the gene expression was detected by reverse transcription polymerase chain reaction (RT-PCR). Results Compared with the sham operation group, the sensitivity of thermal sensation in the model group was decreased (11.23±2.11 vs. 7.14±0.67, P<0.01), and the expression of NGF and its receptor TrkA protein and gene in dorsal root ganglion were significantly down-regulated (0.34±0.05 vs. 0.87±0.11, 0.31±0.04 vs. 0.79±0.10, both P<0.01). After intervention of high-dose ginsenoside Rb1, the thermal pain of the model rats recovered well (P<0.01), with the value of (8.36±0.92); and the protein and gene levels of NGF (0.65±0.07) and TrkA (0.70±0.08) in dorsal root ganglion recovered (both P<0.01). Conclusions Ginsenoside Rb1 can increase the expression of NGF and its receptor TrkA in dorsal root ganglion, maintain the survival of neurons, promote the repair and regeneration of damaged nerves, and improve the sensory function of rats after peripheral nerve injury.
