Abstract:Objective To determine the spa genotyping of foodborne Staphylococcus aureus strains in Fujian Province, to master the genotyping technique and investigate the features of the genotypes so as to provide traceability method and basic data for preventing foodborne disease outbreaks. Methodsspa types of 30 Staphylococcus aureus strains isolated from different kinds of food in food safety risk monitoring in Fujian Province in 2013 were identified by the established polymorphism genotyping method. The X region of spa gene was amplified by PCR and the PCR products were sequenced. The spa types were determined on the official website (http://www.ridom.de/spaserver/). If the type did not have a match in the database, the result of bi-directional sequencing map was uploaded to the official website, confirmed by experts whether it was novel and then given an ID for the new type. Results Among the 30 strains of Staphylococcus aureus, 19 different spa types were identified, including t091 (6 strains), t14534(2 strains), t899(2 strains), t701(2 strains), t1376(2 strains), t189(2 strains), t306(2 strains), t14752, t286, t002, t14475, t4445, t085, t377, t2883, t2310, t796, t127 and t14867. The type of the standard strain (ATCC6538)was t3297. And t14534, t14752 and t14867 were the novel spa genotypes in this study. Conclusions The main spa genotype of the 30 foodborne Staphylococcus aureus strains isolated in Fujian Province in 2013 was t091.
